Not true.  The instrument files (name.exs) should be in the Sampler 
Instruments folder, but the waves thmeselves can be anywhere.

As for basic EXS usage: open the instrument editor (EDIT button), 
create a new Zone.  A strip appears underneath the keyboard, 
representing the zone.  In the lower part of the window a box appears 
containing all the zone parameters.  In this box, click the 
wave/sample-field (or whatever it's called) and pick a sample in the 
fileselector. The low/high key (or whatever *that* is called :-) 
determines the boundaries of the zone.
Create as many zones as you like, line them up across the keyboard, etc...

This should get you going...
