You will lose overall contrast range if you lower brightness hugely, but other than that, just calibrating at the new setting should offer good detail throughout the range. You need reasonable ambient light for a bright display, if you use it in the dark, it will certainly look bad! Run the ambient light feature in Spyder2PRO, if you own that, and choose a white luminance value in its recommended range.
