I've run a Polysix without SSM2044 in five channels before. I don't see why
this shouldn't be possible.
(I did this because the global CV scaling was completely off and it was
easier to measure the tuning for just a single voice - so removing the five
other 2044 and using the Polysix in Unison mode gave me a nice single-voice
synth.)
