If you buy an M-Audio card, you get a restricted version of Ableton Live bundled with it. Live is great and easy to use. I've been so impressed, I shelled out for the full version.
You need an audio card to overcome latency so you can layer things. Otherwise nothing will stay in time. (at least that's my understanding of it)
You need a pre-amp to boost the mic to recording levels
You should record synths directly. Don't mike the amp up - you're just making life difficult for yourself.
