Well, displays work by way of an X-Y matrix - you'd have to have an individually wired cell for each pixel otherwise. So a hole would cut out any action in the affected rows and column.

A custom designed LCD notwithstanding, that would route the pixel lines under the hardware, having something atop the LCD is the way to go :)
