I've previously measured the knob spacing on my synths with a view to  
answering this question. A Sequential Pro-One uses about 20mm between  
the knobs horizontally, and about 40mm vertically, and the Korg  
Polysix is broadly similar.

So, the spacing depends on the knob diameter, but as long as you've  
got 20mm or so between them, it'll probably be ok. Knobs with skirts  
can go a little closer since the body of the knob is narrower than  
the overall diameter.

I also recommend getting a piece of thick cardboard or thin plastic  
or something and just mocking up a dummy panel with a few pots stuck  
through it with knobs on the end. It lets you get a feel for whether  
a given spacing is sufficiently roomy for you or looks at all cramped.
