I use Eagle PCB for single layer. You just don't put any tracks on the
top (or bottom) layer.

I don't think you can get it to autoroute on one layer, but I don't
autoroute anyway.

(Actually, I do occasionally use the top layer - to indicate wire
jumpers where I can't route a track on the other layer).
