You can use a signal generator to drive one primary with a sine wave, then measure the response at the other (IE- using the other primary as a secondary).  You could also doublecheck that you're proposed hookup yields the results you're after, just with scaled-down voltages.  Since it's a power transformer, hitting it with a small AC signal isn't going to hurt anything.
