Depending on the resist you may want to leave it on. I use positive photo 
resist boards from Ever-Muse that I get from Circuit Specialists. They 
recommend leaving the resist on. I was skeptical but it's so thin that 
soldering removes it but the rest of the lands stay coated and it does 
resist tarnishing. It does not interfere with or reduce the quality of the 
soldered connections, which was my main concern.

Another technique if you can't leave the resist on is to use desoldering 
braid, solder and your soldering iron to paint solder onto the lands. This 
is a bit manual and time consuming though.

I'm lazy so I just order my boards with solder coating.
