I've done this before, but understand that the boards did not have
plated-through holes. You would have to have a drill file anyway for
that. I made a transfer to UV film from the printout, and then to the
board. The other layer is similarly done after choosing two targeting
holes to drill (preferably holes placed specifically for this) and align
the other layer.
My boards did not have silkscreen or soldermask (this was a totally
manual operation I did myself in a local circuit board lab), but they
certainly worked well enough to test the design.
